Adverse Reactions
In clinical trials (>10% incidence), arthralgia (10.1%), pain (21.3%), and nausea (8.5%) were most frequent. Other reactions include dizziness (8%), leg cramps (2.6%), and transient hypercalcemia (11% women, 6% men). Post-marketing reports note rare osteosarcoma cases, calciphylaxis (in renal impairment/warfarin users), and hypersensitivity (angioedema, urticaria). Injection site reactions (pain, bruising) and asymptomatic hyperuricemia (3%) may occur. No increased osteosarcoma risk was observed in human studies, but rodent data warrant caution. Drug discontinuation is advised for worsening cutaneous calcification or persistent hypercalcemia symptoms (nausea, lethargy). Renal/hepatic impairment adjustments are unspecified.
